The GMP endonuclease for viral vector manufacturing

In the manufacturing of viral vectors for gene/cell therapies and vaccines, removal of residual DNA and RNA from host cells and plasmids is critical to meet regulatory requirements and streamline vector purification. For this purpose, endonucleases are commonly added to enzymatically cleave polynucleotide chains to smaller fragments of around 3-5 base pairs. DENARASE® is a recombinantly produced endouclease from Serratia marcescens and is widely applied in the industry for the degradation of undesired DNA and RNA.

Working Principle


DENARASE® specifically hydrolyzes the phosphodiester bonds between nucleotides leaving smaller fragments of around 3-5 base pairs. The enzyme is active on all forms of nucleic acids including single-stranded, double-stranded, linear, circular or supercoiled. Best results are obtained when the enzyme is added before the release of DNA.

Production of DENARASE®

DENARASE® has been developed for use in commercial manufacturing processes of biologicals and complies with EU GMP regulations. The patented production process uses a gram-positive Bacillus sp. production host, which minimizes the risks of endotoxins in the product. No antibiotics, materials with TSE/BSE risk or raw materials of animal origin are employed in the manufacture of the product.



DENARASE® vs. Benzonase®

DENARASE® and Benzonase® are both commercial names for a genetically engineered endonuclease from Serratia marcescens. This enzyme degrades all forms of DNA and RNA (single-stranded, double-stranded, linear and circular) without any proteolytic activity. DENARASE® and Benzonase® are the current industry standards due to their efficient activity on diverse DNA/RNA substrates and are widely used in biopharmaceutical production for DNA/RNA removal. Both enzymes are identical in protein sequence and could be applied interchangeably, with the enzyme activity being a key parameter in this context.

DENARASE® - Enzyme Characteristics

Cofactor

DENARASE® uses Mg2+ as a cofactor and a minimum amount is needed for enzyme activity. DENARASE® requires 1- 2 mM Mg2+ cations for optimal activity. A large excess of MgCl2 reduces activity.

Stability and Storage Conditions
DENARASE® is stable within the specification range at a storage temperature of -20 °C for a period of at least 24 months from the date of product release. Note: It is not recommended to store the product at -70 °C or below, since freezing the product will cause loss of activity.

Packaging Information
DENARASE® is filled in non-pyrogenic, USP Class VI-compliant vials. The product vials are packed in polystyrene boxes and shipped under refrigerated conditions. Shipping temperature may differ from storage temperature without affecting product quality.
