Description	SIX1 Reporter Lentivirus: High-quality, lentiviral transcription factor (TF) reporter system that provides a sensitive fluorescent or luminescent readout for human/mouse Sine oculis homeobox homolog 1 (SIX1) transcriptional activity in mammalian cells. The SIX1 reporter is constructed with tandem repeats of the MEF3 site from the Myogenin promoter to readout SIX1 transcriptional activity in mammalian cells. The SIX family of transcription factors function as transcriptional activators or repressors depending on interactions with other highly conserved regulators including activators Paired-box (Pax) and Eyes absent (Eya), repressors Dachshund (Dach) and Groucho (Grg) proteins. Many human SIX gene mutations were located within the SIX domain or homeodomain, suggesting impaired protein-protein interactions or DNA binding activity. The SIX1 binding partner EYA2 enhances SIX1 transcriptional activity. SIX1 and SIX2 hyperactive mutations are associated with Wilms tumor progression. SIX family transcription factors are expressed in different tissues and are key regulators of developmental processes during embryogenesis, and control gene expression to promote self-renewal of progenitor cell populations. The SIX1 reporter lentivirus is purified by PEG precipitation and sucrose gradient centrifugation and is ideal for studying SIX1 transcriptional activity in difficult-to-transfect cells including primary and/or thawed cells.
