Application
Useful as an Ubiquitin ligase in conjunction with E1 and E2 for the study of enzyme kinetics. Ideal for Bir Domain binding and inhibition studies of Caspases 3 7, and 9. Also useful for screening inhibitors, and selectivity profiling.

Ligase Assay Conditions: 50 ul of reaction mix (4 mM ATP, 5mM MgCl2, 1 mM DTT, E1 (0.5 ug), E2(2.8 ug), Biotin-Ubiquitin (0.5 ug) andXIAP (0-1.25 ug) in 50 mM Tris pH 7.4)is incubated for 30 min at 37°C.Transfer the reaction mixture to astreptavidin-coated plate and incubatefor 30 min at 37°C. Add antibody againstXIAP, incubate 1 hr. Then, addsecondary HRP-labeled antibody andincubate 30 min. Finally, add HRPchemiluminescent substrates andread luminescence. Bir Domain Assay Conditions: 50 mM Tris-HCl, pH 7.4, 25 mM NaCl,0.05% Tween-20, 0.01% BSA, 0.5 nM FAM-SMAC peptide. Incubate 30 minutesat room temperature. Fluorescencepolarization is measured at 475 nm/528 nmof exc/em wavelength.
