Reaction wasperformed at room temperature in assaybuffer containing 50 mM MOPS, pH 7.0,60 uM tryptophan, 100 mM ammoniumsulfate, 100 uM ferrous ammonium sulfate,0.5 mM TCEP, 0.3 mM 6-methyltetrahydropterin, 0.05 mg/ml catalase, 1mM DTT and TPH1. Fluorescence signalwas measured at Em360nm/Ex300nm.
Avoidfreeze/thaw cycles. Storing dilutedenzyme is not recommended, ifnecessary, use carrier protein (BSA 0.1- 0.5%).
