MTAP activity wasmeasured using a xanthine oxidase-coupledmethod. MTA cleavage by MTAP produceadenine, which is then converted to 2,8-dihydroxyadenine by xanthine oxidase,leading to an increase in absorbance at 305nm. 50 ul reaction mixtures contained MTAP,50 mM KHsub>2PO4, pH 7.4, 200 uM MTA, 0.02units Xanthine Oxidase, and 1 mM DTT.Activity was measured at 37°C by continuousmonitoring the absorbance at 305 nm for 30min.
