The reactions were conducted at 30°C for 30 minutes in a 25 ul mixture containing 40 mM Tris-HCl, pH 7.4, 20 mM MgCl2, 0.1 mg/ml BSA, various amount of MAT2A enzyme, 0.1 mM L-Methionine, and 10 uM ATP. After enzymatic reactions, 25 ul of Kinase-Glo MAX reagent was added to each well and luminescence was measured using a BioTek Synergy 2 microplate reader.
Avoid freeze/thaw cycles.
