The reactions were conducted at room temperature for 45 minutes in a 25 ul mixture containing 50 mM Tris-HCl, pH 8.0, 10 mM MgCl2, 50 mM KCl, 0.1 mg/ml BSA, various amount of MAT2a enzyme, 200 uM L-Methionine, and 200 uM ATP. After the enzymatic reactions, 50 ul of colorimetric detection reagent was added to each well and the reactions were incubated for another 15 min before the absorbance at 630 nm of each well was measured using a Tecan Infinite M1000 microplate reader.
Avoid freeze/thaw cycles. Protein may be diluted to>=100 ug/ml in PBS + glycerol and stored at -80°C.
