Human recombinant histone tetramer, full length, consisting of 2 molecules each of histones H3 (a.a. 1-136(end)), and H4 (a.a. 1-103(end)), each with an N-terminal His-tag, expressed in an E. coli expression system. GenBank Accession Nos. are NM_003532, and NM_003548, respectively. Global MW = 57 kDa.
Background
The histone tetramer consists of two copies of both histones H3 and H4. It forms the central core of the nucleosome and, as such, plays a prominent role in the assembly, disassembly and positioning of nucleosomes.
Tag
N-terminal His-tag
Formulation
45 mM Tris pH 7.5, 2 M NaCl, 1 mM EDTA, 1 mM DTT and 10% glycerol.
Species
Human
Application
Substrate for histone methyltransferase or acetyltransferase assays. Ideal for screening small molecular inhibitors of histone methyltransferases and acetyltransferases for drug discovery and HTS applications.
Notes
Since the recombinant histone proteins in this tetramer were prepared in E. coli, they will not have methyl, acetyl or many other epigenetic marks. Avoid freeze/thaw cycles.
