Enzyme reactions include 50 ul volumes of 1x Kinase buffer (40 mM Tris pH 7.4, 20 mM MgCl2, 0.1 mg/mL BSA) along with 20 uM ATP, 0.2 mg/mL GSKtidesubstrate, 3mM DTT, and GSTGSK3. After 45 min incubation at30°C, 50uL of the Kinase-Glo reagent solution was added and luminescence from remaining ATP was measured.
