The FcRn (FCGRT/B2M) blocker (Fc-Tag) is a purified human protein corresponding to the Fc region of an antibody which has been engineered to bind to human neonatal Fc Receptor for IgG (FcRn; FCGRT/B2M) with high affinity. This antibody has been tested for specific binding to human FcRn (Biohippo #71283) and it blocks the binding of FcRn with immunoglobulins G.

Assay 1. A 96-well plate was coated overnight at 4°C with 50 ul/well of the FcRn blocker at a concentration of 2 ug/ml, diluted in PBS. The plate was washed 3 times with Immuno Buffer 1 (Biohippo #79311) and blocked with 100 ul of Blocking Buffer 6 for an hour at room temperature. A serial dilution of biotinylated FcRn protein (Biohippo #71283) prepared in Blocking Buffer 6 was added for 30 minutes. After 3 washes, the plate was incubated with Streptavidin-HRP followed by more washes and addition of a colorimetric HRP substrate. After quenching, color intensity was measured at 450 nm.Assay 2. The effect of FcRn (FCGRT/B2M) Blocker was assessed using the Fc(IgG1):FcRn Inhibitor Screening colorimetric kit (Biohippo #78501) using the following conditions: Fc(IgG1) protein at 2 ug/ml, diluted in PBS, serial dilution of FcRn Blocker prepared in blocking buffer, and 1.5 ug/ml of FcRn-Biotin.
