RPMI-1640 represents an excellent cell culture medium for human and animal lymphocytes, either normal or neoplastic. Murine myeloma cells proliferate in this medium as well, if it is supplemented with 15% serum. Selection media based on RPMI 1640 for the generation of hybridoma are supplemented with HAT or HT. The RPMI media were developed at the Roswell Park Memorial Institute, Buffalo, N.Y., USA and meanwhile a variety of culture media based on RPMI are in use.
This basic RPMI 1640 High glucose must be supplemented with serum or hPL to promote cell proliferation.
Maintenance
Immediately after delivery, store the medium at 4°C to 8° C in the dark. Do not heat the medium over 37°C or use uncontrollable sources of heat (e.g. microwave appliances). If only a part of the medium is to be used, remove this amount from the bottle and warm it up. Do not freeze the medium. This can result in high salt concentrations by freezing out pure water which will cause irreversible damage.
