This medium was named for the site at which it was developed, the Roswell Park Memorial Institute. RPMI 1640 medium represents a modified McCoy’s 5a medium with an increased level of inositol. A variety of cell types such as mammalian myeloma and hybridoma cells, human leukocytes, and B and T lymphocytes proliferate well in this medium when supplemented with serum. RPMI 1640 is also suitable for suspension cultures and human leukemic cells.
RPMI 1640 represents an excellent cell culture medium for human and animal lymphocytes, either normal or neoplastic. Murine myeloma cells proliferate in this medium as well, if it is supplemented with 15% serum. Selection media based on RPMI 1640 for the generation of hybridoma are supplemented with HAT or HT. The RPMI media were developed at the Roswell Park Memorial Institute, Buffalo, N.Y., USA and meanwhile a variety of culture media based on RPMI are in use.
pH = 7.2 +/- 0.2. Each lot of RPMI 1640 (CLS designation MG-70) has been tested for sterility and absence of Mycoplasma and Bacteria. The manufacturing process and documentation are performed according to ISO9001:2008 based on GLP under strict quality control.
Maintenance
Keep refrigerated at +2°C to +8°C in the dark. Freezing as well as warming up to +37°C minimize the quality of the product.
Do not heat the medium to more than 37°C or use uncontrollable sources of heat (e.g. microwave appliances).
If only a part of the medium is to be used, remove this amount from the bottle and warm it up inside a separate device.
