This enzyme is necessary for target cell lysis in cell-mediated immune responses. It cleaves after Asp. Seems to be linked to an activation cascade of caspases (aspartate-specific cysteine proteases) responsible for apoptosis execution. Cleaves caspase-3, -7, -9 and 10 to give rise to active enzymes mediating apoptosis.
Specificity
Natural and recombinant Mouse Granzyme B(G,H)
Subcellular Location
Cytoplasmic granule Cytoplasmic granules of cytolytic T-lymphocytes and natural killer cells.
Detection Range
15.6-1000pg/mL
Entryname
GRAB_MOUSE
Linearity
The linearity of the kit was assayed by testing samples spiked with appropriate concentration of Granzyme B and their serial dilutions. The results were demonstrated by the percentage of calculated concentration to the expected.
Intra-assay Precision (Precision within an assay):Three samples of known concentration were tested twenty times on one plate to assess intra-assay precision.
Intra-Assay CV: ≤4.1
Inter-assay Precision (Precision between assays):Three samples of known concentration were tested in five separate assays to assess inter-assay precision.
Inter-Assay CV: ≤6.9
Protein Name
Granzyme B(G,H)
Recovery
Recovery was determined by spiking various levels of Granzyme B into serum and plasma .
