The predicted PRTFDC1 protein contains a phosphoribosyl transferase domain, but it lacks conservation of 11 HPRT1 residues critical for HPRT function, suggesting that PRTFDC1 has lost its ancestral HPRT activity. Database analysis revealed that Prtfdc1 is present in several vertebrate species. However, in mouse, the Prtfdc1 gene appeared to be inactive due to 3 independent mutations.
PRTFDC1was inactivated either by deletion or hypermethylation in a significant subset of oral squamous cell carcinomas. Restoration of PRTFDC1 expression in 1 of these lines inhibited cell growth in colony formation assays, whereas knockdown of PRTFDC1 expression in PRTFDC1-expressing cells promoted cell growth.
