TNK1 is a nonreceptor tyrosine kinase. These kinases, like members of the SRC and JAK families, mediate intracellular signaling downstream of receptor activation.
TNK1 contains a putative N-terminal phosphotyrosine-binding domain, followed by the tyrosine kinase domain, a putative SH3 domain, and a proline-rich C terminus that contains SH3-binding motifs. The kinase domain has all 12 structural features conserved in tyrosine kinases, including an ATP-binding site. TNK1 shares highest homology with ACK, including 45% amino acid identity over a 413-amino acid span that includes the kinase domain and putative SH3 domain. specific expression of 4 transcripts between 1.0 and 9.5 kb in fetal brain, lung, liver, and kidney.
