A Vector Map
- 1. The name and size of the vector are indicated in the middle of the circular (5299 bp)
- 2. The elements of a vector
- -The origin of replication site Ori, the site that controls the origin of replication
- -Antibiotic resistance genes, such as Amp+, Kan+
- -Multi-cloning site MCS clones, carry foreign gene fragments
- -P/E promoter/enhancer
- -Terms, the termination signal
- -Adding poly(A) signal to stabilize mRNA
ITR: the viral origin of replication and packaging signal.
Promoter: hSyn (human Synapsin I) promoter.
GOI/shRNA: hChR2(H134R).
Commonly used tags: mCherry, red fluorescent protein.
WPRE: Woodchuck hepatitis virus post-transcriptional regulatory element, generally placed after the CDS of the vector and before polyA, can help stabilize mRNA and improve the expression of the target gene.
polyA: Poly A tail, used to terminate transcription.
rAAV2/9: A shell with the ITR of AAV2 and AAV9.
Promoter: hSyn (human Synapsin I) promoter.
GOI/shRNA: hChR2(H134R).

Shipping and Storage Conditions
1. Shipping: AAV viruses are shipped with dry ice.2. Storage: Store AAV viruses at -80°C. Avoid repeated freeze-thaw cycles by storing in small aliquots.
3. Subpackage: If the amount required for each experiment is small, please dispense a small amount of melted AAV into multiple tubes immediately after receiving the product, and then store it at -80 ℃. If needed, AAV can be diluted with sterile PBS buffer (currently equipped and used).
Biosafety
Recombinant AAV constructs do not encode for either a potentially tumorigenic gene product or a toxin molecule. According to guidelines from the National Institutes of Health (NIH), recombinant AAV vectors can be handled in a Biosafety Level 1 (BSL-1).
