Key Features of the ATHENA II Platform

Random Peptide Insert Library:

The ATHENA II platform utilizes random peptide insertion into variable sites of the AAV capsid protein to create a highly diverse library. These libraries allow for the identification of novel capsids that exhibit unique properties, such as altered tissue tropism, enhanced transduction efficiency, and lower immune responses, which are critical for gene therapy applications.

Premade AAV Capsid Libraries (II)

 AAVnerGene offer premade AAV Capsid Libraries (II) where the CAP gene with random peptides is driven by a CAG/P40 hybrid promoter. The P40 promoter is used during AAV production, while the CAG promoter is used for CAP expression in target cells or tissues. Most premade libraries insert peptides at the VR-VIII and VR-IV regions, which are known for their roles in capsid function.

2.          AAV library production. The AAV library plasmid was transfected as 200 Copies/cells together with our mini-pHelper-Rep.
3.          AAV purification: two rounds of CsCl ultracentrifuge.
4.          For the premade AAV capsid library, we only provide AAV virus but not the plasmid.
