ATHENA II AAV Capsid Platform
The ATHENA-II platform is a state-of-the-art random peptide (RP) insertion AAV capsid library system designed to develop novel AAV capsids with tissue-specific tropism. In these AAV capsid libraries, random peptide sequences are inserted into specific variable regions (VRs) of the AAV capsid protein. These modifications can significantly alter the capsid's properties, such as its tissue tropism, transduction efficiency, and immunogenicity, making them more suitable for various gene therapy applications. By screening these libraries, researchers can discover and select novel AAV capsids with enhanced properties that are ideal for specific therapeutic needs.
Biohippo, in partnership with AAVnerGene, offers high-complexity AAV capsid libraries containing over 1 billion unique variants, providing a vast resource for exploring new capsid functionalities. We also provide custom AAV capsid library construction services, allowing researchers to tailor their libraries by choosing specific templates, random peptide insertion sites, variable regions, and desired levels of complexity.
ATHENA II Capsid Library Kits
In partnership with AAVnerGene, we offer premade AAV Capsid Libraries (II) where the CAP gene with random peptides is driven by a CAG/P40 hybrid promoter. The P40 promoter is used during AAV production, while the CAG promoter is used for CAP expression in target cells or tissues. Most premade libraries insert peptides at the VR-VIII and VR-IV regions, which are known for their roles in capsid function.

1. Complexity. The complexity was calculated according the numbers of clones after electroporation.
2. AAV library production. The AAV library plasmid was transfected as 200 Copies/cells together with our mini-pHelper-Rep.
3. AAV purification: two rounds of CsCl ultracentrifuge.
4. For the premade AAV capsid library, we only provide AAV virus but not the plasmid.
