The determination of hydroxyproline in skin is used as a screening index of anti-aging medicines. The excretion of hydroxyproline will increase with age in development stage, then keep stable after non-development. The hydroxyproline in elderly is less than the young. When rats suffer from spontaneous hypertension and secondary hypertension, the content of hydroxyproline will increase in their urine. Surgical trauma will result in an increase of hydroxyproline in the urinary excretion. High lipid, low protein, alcoholic poisoning toxicant, malnutrition, or liver cell degeneration and necrosis will lead to the increase of fiber by inflammation which splits liver lobule, leading to cirrhosis of the liver. When the liver fibrosis, the collagen fibers are the mainly increasing composition in liver, which uniquely have the hydroxyproline. The content of liver hydroxyproline can be converted to the content of the liver collagen, reflecting the degree of liver fibrosis. The decomposition of collagen fiber mainly depends on collagen enzyme and other protease. The hydroxyproline decomposed products will excrete by the urine. The collagen degradation can be reflected by the content of urinary hydroxyproline. By determining the content of hydroxyproline in the tissue and urine, the degree of fibrosis can be judged, and the preventing and treating drugs can also be screened.
