Description	p53 is a tumor suppressor protein that plays a large role in the cellular response to DNA damage and other genomic aberrations. p53 activation can lead to cell cycle arrest and DNA repair or to apoptosis (1). p53 phosphorylation occurs at multiple sites in vivo, and p53 can be phosphorylated by several protein kinases in vitro (2). p53 phosphorylation at Ser15 and Ser20 is induced by DNA damage, leading to reduced interaction between p53 and its negative regulator, oncoprotein MDM2 (3). MDM2 targets p53 for ubiquitination and degradation (4). ATM, ATR, and DNA-PK can phosphorylate p53 at Ser15 and Ser37, while Chk2 and Chk1 can phosphorylate p53 at Ser20 (5). MDM2 does not strongly bind phospho p53, causing phospho p53 accumulation and activation in response to DNA damage (3).
