Description	This AAV expresses hGtACR1-P2A-EGFP driven by an ubiquitous Ef1alpha promoter in Flippase (FLP)-dependent manner. The 1.2 Kb EF1alpha promoter is a commonly used, constitutive promoter derived from human elongation factor-1 alpha (EF1alpha). It is often used in conditions where the other constitutive promoters, such as CMV, have diminished activity or have been silenced. FDIO is Flp-dependent Double-Floxed Inverted Open reading frame (fDIO) cassette. hGtACR1 is human codon-adapted GtACR1. GtACR1 (Guillardia theta anion channel rhodopsin 1) is a light-gated anion channel rhodopsin derived from cryptophyte algae Guillardia theta. Its channel properties determine that it is more efficient than the commonly used NpHR. Anion channel rhodopsins (ACRs) provide highly sensitive and efficient membrane hyperpolarization and neuronal silencing through light-gated chloride conduction. The optimal wavelength for GtACR1 activation is 515nm. The short 2A peptide sequences, when cloned in-frame between two genes, allow for efficient, stoichiometric production of discrete protein products within a single vector through a novel "cleavage" event within the 2A peptide sequence. This differs from conventional approaches for multiple protein expressions, such as IRES-mediated bicistronic gene expression, which has several limitations including imbalanced protein expression. The use of 2A peptide sequences alleviates these concerns, since 2A-mediated "self-cleavage" gives rise to a 1:1 ratio of the two separate proteins. Several forms of 2A peptide are commonly used: T2A (Thoseaasigna virus 2A), P2A (porcine teschovirus-1 2A), E2A (equine rhinitis A virus), and F2A (foot-and-mouth disease virus, FMDV 2A). EGFP is a basic (constitutively fluorescent) green fluorescent protein published in 1996, derived from Aequorea victoria, with a Excitation/Emission spectrum around 488nm/507nm. It is reported to be a rapidly-maturing weak dimer with moderate acid sensitivity.
