Hepatitis C virus (HCV) genotype 1b, also known as HCV1b, is a fusion protein of a.a. 21-32 of NS4A, linker (4 a.a.) and a.a. 3-181 of NS3 (GenBank Accession No. AF054247) with Asp-to-Val mutation on a.a. 168, and N-terminal FLAG-His-tag, MW = 22.3 kDa, expressed in an E. coli expression system.

Assay was performed in 100 ul HCV assay buffer containing 50 mM Tris, pH 7.4, 150 mM NaCl, 5 mM DTT, 10% glycerol, and 5 uM fluorescently labeled HCV substrate peptide. Reaction was continuously monitored at room temperature for 20 min. at ex350/em500.
