Fusing a kinase domain to a homodimerization domain such as the PNT domain of ETV6 can result in activated kinase. The NTRK3 Stable Ba/F3 Cell Line expresses activated NTRK3 tyrosine kinase that is linked to a GFP reporter (GFP-ETV6-NTRK3 in SED2 vector). Ba/F3 cells expressing activated tyrosine kinase become completely dependent on the kinase activity for survival and as a result, the cells can be propagated even in the absence of IL-3. NTRK3 belongs to the neurotrophic tyrosine receptor kinase (NTRK) family consisting of proteins which bind to neurotrophins. Protein members of this family are crucial in embryonic development by assisting with the formation of the central and peripheral nervous systems. They are also involved in the maturation of these two systems. NTRK3 is a membrane-bound receptor that, upon neurotrophin binding, phosphorylates itself and members of the MAPK pathway. Signaling through this kinase leads to cell differentiation and may play a role in the development of proprioceptive neurons that sense body position. Mutations in its gene have been associated with medulloblastomas, secretory breast carcinomas and other cancers.
