Fusing a kinase domain to a homodimerization domain such as the PNT domain of ETV6 can result in activated kinase. The KDR Stable Ba/F3 Cell Line expresses activated KDR tyrosine kinase that is linked to a GFP reporter (GFP-ETV6-KDR). KDR belongs to the VEGF receptor tyrosine kinase family consisting of proteins which bind to vascular endothelial growth factors. These proteins play an important role in forming the vascular system during embryogenesis and beyond, in conjunction with maintenance. KDR functions as the main mediator of VEGF-induced endothelial proliferation, survival, migration, tubular morphogenesis and sprouting. The signaling and trafficking of this receptor are regulated by multiple factors. Mutations of its gene are implicated in infantile capillary hemangiomas.

The base medium for this cell line is Prigrow II medium available at abm, Cat. No. TM002. To make the complete growth medium, add the following components to the base medium: fetal bovine serum (TM999)* to a final concentration of 10%, 10 ng/ml mouse IL-3 (Z200145), 1mM HEPES (Corning), 1% GlutaMAX (Gibco), 1% Non-essential Amino Acids (Corning), 1mM Sodium Pyruvate Solution (TM057) and Penicillin/Streptomycin Solution (G255) to a final concentration of 1%. Once the cells have recovered from thaw 1 µg/ml Puromycin (G264) can be added.Change media every 2-3 days.Carbon dioxide (CO2): 5%, Temperature: 37.0°C.* Do not use heat-inactivated FBS for cell culture unless specified otherwise. Use 30% FBS for the initial recovery of these cells post thaw, reduce to 10% FBS once the cells have recovered.
