GNAO1 R243H Stably Expressing Immortalized Human Mammary Epithelial Cell line expresses GNAO1, the protein of which represent the alpha subunit of the G0 heterotrimeric G-protein signal-transducing complex and is partially responsible for smooth muscle contraction and depressing neuronal activity. This genes role is to inhibit adenylate cyclase , open K -channels (via beta/gamma subunits), and close Ca2 -channels. This particular cell line expresses the the Arginine243?? Histidine (R243H) mutation of GNAO1, which has been identified in breast carcinomas and is known to promote oncogenesis when introduced into cells. The oncogenic R234H mutation renders Galphao constitutively active by accelerating the rate of nucleotide exchange, which enhances signaling pathways that are associated with neoplasmic transformation. This cell line was made using retrovirus transduction. It carries flag tag and expresses GFP.
